Epigenetic changes to chromatin include DNA methylation and histone post-translational modifications. These marks were initially classified as either activators or repressors of gene transcription. In this way, simple combinations of marks would have predictable transcriptional outcomes. Evidence now shows that some marks have opposing roles depending on their context, so chromatin marks represent a more complex 'language' than has been appreciated previously.
