Betel nut contains alcaloids such as arecoline, guvacoline, and guvacine known as M1, M2, and M3 muscarinic receptor agonists. Meanwhile, pupil size is controlled by two muscles, i.e., sphincter pupillae musculus that has M3 muscarinic receptor, and dilator pupillae musculus that has a1 adrenergic receptor. The activation of the M3 muscarinic receptor at sphincter pupillae musculus by its agonists (e.g., betel nut alcaloids) leads to contraction of that muscle which results in miosis. This research was conducted to prove the ability of betel nut decoction (Areca catechu) in causing miosis on the rabbit eyes (Lepus sp.) through topical administration on that rabbit eyes. This research was an experimental study using pretest-postest design method. It used five rabbits as the experimentation animals. On each rabbit was administered four doses of betel nut decoc solution, i.e., 0.0001%, 0.001%, 0.01%, and 0.1%. The main parameter was rabbit eyes pupil diameter before and after each solution was administered. The result showed that the rabbit with all doses of the solution gave significant smaller pupil diameters than controls (Friedman; p = 0.0006; a = 0.05/p < 0.05). There was a good coffelation between increasing doses of the solution with the smallest diameter achieved on each dose (Spearman, r = 0.713), eventhough the diameters of the doses are not significantly different (Wilcoxon; p > 0.05; a = 0.05). It can be concluded that betel nut decoction may be used as mioticum.
