Proving ethnomedicinal information of Gendarussa vulgaris Nees. as male contraceptive is one of developing effort of Indonesian traditional drugs. Some researches have been done on various extracts or fraction of G.vulgaris leaf, all to reveal the mechanism of action of active compounds against animal beside elucidating its active compounds. In this research, n-butanol fraction that known to contain flavonoid (polyphenol) by HPLC-DAD was used . Male mice was used as experimental animals, doses with 15 mg/20 g b.w. and 30 mg/20 g b.w. p.o. once a day for 55 days. Decrease of hyaluronidase activity of mice spermatozoa was shown by inhibition of mice spermatozoa penetration in vitro fertilization process after 7 hours incubation. Result shown that administration of the polyphenol fraction, could inhibition mice spermatozoa penetration IVF shown by no zygote developed. Further more also put identified that no dispersion of cumulus oophorus or granulose cell.
