The objective of this experimental were to obtain the effective genetic transformation procedures on soybean NYI1I and GUS genes on KIWI plasmid vector carried by Agrobacterium transformed into three soybean varieties, Experiment were set up the effect of (a) Agrobacterium concentration, (b) time of preculture, (c) time of Agrobacterium inoculation, and (d) time of preculture to effective transformation process on three soybean varieties. Meanwhile, in vitro regeneration of transgenic tissues was conducted too. So that can be as model to transfer target genes on soybean Soybean tissue can be transformed by foreign genes, with Agrobacterium concentration O.5x 1 07 cells/ ml media. (OD600 nm.), preculture time 5-10 days, cocu1tivation Lime 2-3 days. Time of Agrobacterium inoculation non sigi1if.cani in effecting transformation process: Immature cotyledon2 was transformed conferred somatic embryos and embryo seedlings kanamycine resistant.
