Fighting chemotherapy-resistant cancers remains one of the most challenging tasks in the clinic, and the battle is often lost. Artificial nucleoside or nucleobase analogues (antimetabolites) constitute first-line therapy against many hematologic cancers and certain solid cancers. Initial rates of response to these treatments are often high, but adverse effects are dose-limiting and alterations in the nucleotide-metabolism pathways can result in drug resistance.
