Cancers arise as a result of mutations that cause unbridled growth by deregulation of the normal cell cycle. A case in point is grade II or III gliomas, the large majority of which express a single mutation in the gene encoding isocitrate dehydrogenase 1 (IDH1). This substitution mutation (R132H) is considered to be a root cause of the initiation of these tumors.
