A research on isolation and characterization of phenol degraded bacteria isolated from an oil well in Sumatera has been done. Three kinds of bacteria for bacterial isolation from crude oil and formation water samples : Stone Mineral Salts Solution (SM), Bushnell Haas Mineral Salts (BH), and Leedbetter's Medium (LB). Eight bacteria isolated were obtained and only one has the capability to selected for further research. The growth was optimize at inoculum concentration of 10%. Isolate was culture for 8 days at 50 Celcius Degree, 120 rpm, then the growth curve was made. Degradation of phenol was observed from bacterial culture in SM containing 1% phenol. Based on the optimization results, the optimal growth conditions were SM medium containing o.1% years extract, with initial PH = 7, and inoculum concentration of 10%. HPLC analysis for phenol showed that the phenol value declined about 59,211%.
