The Wharton’s jelly-derived mesenchymal stem cell (WJ-MSCs) from the umbilical cord has been shown to have faster proliferation rates and greater expansion capability compared with others adult MSCs. In this work, the isolation and culture of WJ-MSCs using fetal bovine serum (FBS) as supplement was investigated in term of growth kinetics, differentiation capacity, and cell surface markers. WJ-MSCs of 3 donors were culturedin MEM Alpha medium supplemented with 20% FBS. Growth kinetics, cell surface markers, and in vitro differentiation potential toward the adipogenic, chondrogenic and osteogenic lineage were investigated. The study demonstrated that WJ-MSCs can be isolated and cultured in MEMAlpha medium supplemented with 20% FBS. Differentiation capacity was preserved throughout long-term culture. Immunophenotype assay showed similar MSCs characteristics with the exception of a distinct CD14-positive side population. However the CD14-positive side population tends to diminish with prolonged culture. All these parameters matched quite well with MSCs characteristics. Thus we established an efficient protocol for propagation of human WJ-MSCs. protocol for propagation of human WJ-MSCs.
