Conserved gen such as gen coding rRNA (ribosal RNA) has been used extensively to analyze microbial phylogenetic relation in taxon category. Phylogeny tree that is obtained as phylogenetic classifisation result showed phylogenetic relation for bacterial strains that is classified. The value of DNA-DNA relatedness has been used as the basic for defining microbial species as a term genomic species concept. this research was aimed to introduce molecular taxonomy procedure, especially phylogenetic classification based on molecular data in the form of gen coding 16s rRNA (16S rDNA) for bacterial strains that was classified, including bacterial strains of genus Bacillus. For constructing phylogeny tree that was based on 16s rRNA sequences, it had been done some research stage were: preparation of 16S rRNA sequences, alignment of 16S rRNA sequences, constructing of phylogeny tree, visualization of phylogeny tree, editing phylogeny tree, presentation of phylogeny tree, and constructing of similarity matrix and the difference of 16S rRNA nucleotides. From this research was obtained that based on genomic species concept, 20 bacterial strains of genus Basillus that was tested included in one cluster or one species group, because of all strains had similarity value = 70%. Phylogeny tree and similarity value showed evolutionary phylogeny relation interbacterial strains and describe closer of genetic relation interbacterial strains was tested.
