Abstract
Tempe, an Indonesian fermented food, is widely consumed as a nutritious protein source. The quality of Indonesian tempe is highly determined by the presence of various bacteria during the fermentation process. One genus of important bacteria in tempe is Klebsiella because its ability to produce vitamin B12. The content of vitamin B12 in tempe is a unique feature because this vitamin is rarely found in plant-based food. The objective of this research is to compare genetic diversity of Klebsiella in tempe based on their 16S rRNA encoding gene. Tempe samples were obtained from several producers tempe in Bogor and Jakarta area. MacConkey Agar was used as medium to culture Klebsiella. Genomic DNAs were extracted using Genomic Purification Kit, and 3730XL DNA sequencer was used to sequence of 16S rRNA gene. The amplification of the 16S rRNA encoding gene region was carried out using 63f (5'-CAG GCC TAA CAC ATG CAA GTC-3') and 1387r (5'-GGG CGC WGT GTA CAA GGC-3’) primers. Phylogenetic tree was construded with a 100 bootstrap support neighbour joining method using Molecular Evolutionary Genetics Analysis (MEGA 4). Based on biochemical tests, we obtained 23 suspeded of Klebsiella isolates out of 111 bacterial isolates from 14 different tempe samples. Based on 16S rRNA gene sequence, it was found that they consisted of three Enterobacter species and 20 Klebsiella species. comprising of K. pneumoniae (6 isolates), K variicola (6 isolates), dan Klebsiella sp. (8 isolates). The value of maximum identity ranged from 98-100%, except for two isolates that were 91% and 93% similar to the data base. These 20 isolates were phylogenetically clustered into six groups. This result demonstrated that the Klebsiella species present in tempe are genetically diversed.
