Abstract
Introduction
Low bone mineral density may reflect osteoporosis, and bone mineral density can be influenced by the lifestyle and genetic factors. Since Vitamin D Receptor (VDR) gene plays an important role in bone mass regulation, polymorphism in VDR gene has been suggested to associate with osteoporosis. The aim of this cross-sectional study was to investigate the influence of lifestyle factors on bone mineral density and to relate with the VDR gene polymorphism in Chinese population living in Jakarta.
Methods
Bone mineral density of Chinese ethnics (n 118, aged 35-90 year) was measured by dual-energy X-ray absorbtiometry and quantitative ultrasound calcaneous densitometry. t-score was measured to indicate normal, osteopenia and osteoporosis state. Lifestyle information was obtained by questionnaire. Genomic DNA was isolated from peripheral leucocytes and VDR polymorphism was determined by PCR-RFLP using restriction enzyme Bsm1.
Result
Bone mineral density was associated with older age (p<0.001). Further analysis, bone mineral density was positively correlated with the amount of coffee consumption and duration of exercise with Pearson correlation value of 0.187 and 0.173, respectively. The frequencies of the VDR Bsm1 polymorphism in the participant were 11.02% and 45.76% for Bb and bb, and the proportions of Bb and bb to osteoporosis were 4.5%, and 26.9% respectively, indicating that the VDR polymorphism was positively correlated to bone mineral density.
Conclusion
The lifestyle, especially exercise and the period of coffee consumed, and age of the respondents play significant role on the incidence of osteoporosis. The genetic background of VDR gene polymorphism Bsm1 might be useful for identifying individuals at risk for osteoporosis. Detection of bb VDR polymorphism might be of interest to early change of the lifestyle.
