In this application note, we demonstrate the capabilities of the BD Pathway™ high-content bioimager and associated software for imaging and analyzing various cellular microbiology assays that target different stages in the life cycle of an intracellular pathogen. The J774A.1 mouse-macrophage cell line and L. monocytogenes were used as a model system to conduct multiwell plate-based, endpoint (fixed-cell) assays for phagocytosis, phagosomal escape, and intracellular replication. In addition, we used labeled Escherichia coli particles to demonstrate the utility of live-cell, kinetic assays to study hostpathogen biology. |
