Abstract
Two mosquitocidal toxins (Mtxl and Mtx2) of Bacillus sphaericus produced during vegetative growth were considered as potential candidates for vector control due to their toxicity against some mosquito larvae. High cost fennentation for growing these bacteria and narrow host range comparable to chemical insecticides were some reasons limiting the usage of B. spizoericus as vector control. Combination of the genes (Mtxl and Mtx2) and cloning it into novel host (E. coil) could be applied in order to increase the toxicity and broaden the host range. To increase the expression of Mtxl and Mtx2, both proteins were designed as a fusion protein with Gluthathione-S-Transferase (GST). To construct the recombinant plasniid (pGEX-tMtxl-Mtx2), GST-Mtx2 gene was subcloned into pGEX-tMtxl and subsequently transformed into E. coil JM1O9. Protein expression from this recombinant plasmid was assayed against Aedes aegypli larvae. Both toxins (GST-tMtxl and GST-Mtx2) were successfully co-expressed in E. coil JM1O9 even though their expressions were not as high as the controls that were only expressing single toxin. From larvicidal activity assay, E. coli that was expressing both toxins showed higher toxicity (LC50of about 104-105 cells/mi) compared to E. coli expressing only a single toxin (LC50 of about 105~106cells/mi). This result indicated synergism activity between GST-tMtxt and GSTMtx2
