Isi artikelObjective: The aim of this study is to study the profile of human spermatozoa membrane protein based on identification of molecular weight, protein concentration, phosphorylation activity, and antibody production. Material and method: There are three phases in this study. First, isolation of human spermatozoa membrane protein using a lysis buffer containing Tween-20. Second, determination of molekular wight, phosphorylation activity, and protein concentration. Third, determination of antibody production. Molecular weight of protein membrane was determined by SDS-PAGES, confirmation of protein concentration as measured by Biuret method, kinase activity was measured by spectrophometry at its optimum condition, and production of antibodies were confirmed by ELISA. Result: Human spermatozoa contained proteins with molecular weight of 201, 163, 116, 97, 72, 56, 48, 32, 27, 20, and 17 kda. The highest phosphorylation activity was found in the protein with molecular weight of 20kda, 30,197 x 10 pangkat-3 unit. Lowest phosphorylation activity of 28,976 x 10 pangkat -3 unit was shown in protein with 116 kda of molecular weight. This protein was shown to induce production of antibodies. Biuret method showed the protein concentration of 116 kda protein of 410 miug/mL. Conclusion: The protein with molecular weight of 116 kda can be used as an immunocontraception agent because of lower phosphorylation activity, good concentration, and ability to iduce production of antibodies.
