Abstract
Dengue infection is highly endemic in Indonesia with increasing incidence rate every year.
Dengue treatments are usually given symptomatically and essential to increase the patient’s survival rate if given in early phase of infection. Accurate and rapid dengue virus detection is important to diagnose and treat dengue patient. A multiplex assay (Luminex technology) is
developed to detect dengue viruses. Monoclonal antibody from mouse ascitic fluid and hybridoma
supernatant were produced and used in this study. It was shown that incomplete Freund’s adjuvant
(IFA) gave better ascites production compared to pristane. The usage of higher hybridoma cells
injected also gave higher volume of ascites. However, an optimum injection condition has to be
generalized for each hybridoma. Luminex assay using 2.5 µg of 3H5 monoclonal antibody could
detect 5x103 PFU dengue-2 (DEN-2) virus stock while 2.5 µg of 1H10 monoclonal antibody could
detect 75 PFU dengue-4 (DEN-4) virus stock. Further coupled antibody and reporter optimization
needs to be done to increase the sensitivity of DEN-2 and DEN-4 detection. The usage and
optimization of anti-DEN-1 and anti-DEN-3 monoclonal antibody needs to be done in order to
develop a multiplex assay for dengue diagnosis. Luminex assay is an alternative laboratory
technique to diagnose dengue infection rapidly and cost-effectively.
