Abstract
Aquilaria malaccensis Lank. is the plant that has a high economic value since its wood can produce resin called gaharu if it is infected with certain fungi, and gaharu is smell very good. Gaharu volume exported from Indonesia year after year is keep increasing. How ever, in the recent years its volume is lowered down 30 tons which is valued about US $ 600.000. Unfortunately almost all of gaharu just directly harvest from the forest. This phenomena can reduce plant population in Indonesia and may cause it rarely exist. Suppoerted technology such as plant propagation technique and inoculation of spesific fungi for a good quality gaharu is not available yet. Based on property and usage of gaharu, Aquilaria malaccensis Lank. is classified to has a high rank to be develop. One technology that can be employed to develop this plant population is in vitro culture technique. Propagation through in vitro culture technique can be done by adventive shoot growth, lateral shoot proliferation and somatic embriogenesis. Commonly, woody plant is hardly to be profilerated and generated, therefore plant growth regulator manipulation is needed to overcome those problems. Either cytokinin alone or in combination with auxin can be used in the media formulation to produce the whole plant. Experiment which have been done this year had an objective to get 1) initiation method, 2) shoot multiplication , and3) embriogenic callus induction through in vitro culture. Problem that need to be solved for induction and shoot multiplication were sterilization method and reducing browning occurrence. In this stage, several types and time duration for sterilization have been employed and also usage of several antioxidants. In order to stimulate shoot initiation, sterile juvenile axilar shoot have been planted on either MS media, Anderson or WPM which each of them contain in accordingly BA 1 – 5 mg/l. SUbculturing has been done several times in order to increase multiplication rate and shoot juvenilization. Mean awhile for somatic callus induction, those three media mentioned above also have been used but instead it using either Picloram or 2,4 D with accordingly 1, 5 and 10 mg/l consentration. Result showed that combination of alkohol 70% in 3 minutes, HgCL2 0,2g/l in 2 minutes, clorox 30% in 5 minutes, and clorox 20% in 5 minutes, gave 50% success of sterilization process, and usage of antioxidant can overcome browning peoblem. Capacity of shoot juvenilization and multiplication can be increased after subculturing to a new media. Addition of BA 3 mg/l on the basic media in avarage can resulted 7 shoots on WPM.
