Abstract
Microsatellite is molecular marker base on sequence DNA that has been used for
many genetic diversity analyses on plants. The advantages of this molecular
marker, for instance high polymorphism and spread randomly across the genome
have made this marker a choice for genetic diversity study and heterozygosity
among mango varieties. Some microsatellite motif have been discovered from
mango i.e. GT24AG10, T7ATGT3AT3GT17, AAG4TCC3AAT3, and CCG6. Primer
pairs also have been designed and synthesized so they can flank the tandem
repeat sequence. By using touch down PCR protocol, polymorphism DNA bands
have been successfully amplify from mango genome. This was strengthening by
using PAGE and silver staining. PIC (Polymorphic Information Content) value that
show an expected heterozygosity and distinguishing strength among the PCR
primer pair, give a high value (above 0.5). This shown that there is diversity among
mango varieties.
