Abstract
This study was performed to test the affects of growth media on inositol uptake and utilization in C. neoformans. When C. neoformans C21 cells were grown in media containing 2% mannose (MOM), 2% xylose (MOX), or 2% glucuronic acid (MOGA) inositol transport was biphasic. There were two transport components detected, a high-affinity (low K$/sb[/rm m]$) and a low-affinity (high K$/sb[/rm m]$) transport component. The intracellular inositol concentration was evaluated in C. neoformans C21 cells grown in MOGA, MOM and MOX using High Performance Liquid Chromatography (HPLC). The intracellular inositol concentration was significantly (p = 0.002) lower for cells grown in MOX. Utilization or inositol for lipid or carbohydrate metabolism was examined in C21 and C20 cells grown in media containing 2% inositol (MOI) or 2% glucose (MOG). The distribution of radiolabeled inositol in both phosphatidylinositol (PI) and the carbohydrates inositol and glucuronic acid was not affected by the growth media. A PCR probe was generated and used to screen a $/lambda$ZapII C. neoformans cDNA library. A putative clone 1.2 kb was isolated. It shared 39-42% homology with other inositol transporters that have been cloned and sequenced. These studies contribute to understanding the pathogenicity of C. neoformans.
