Synthesis study of Polylactide base biodegradable nanosphere has been done. Nanosphere was synthesized via emulsion evaporation process. Emulsion process was done using ultrasonic irradiation. Sonication times were varied for 2, 4, 6, 8, 10 and 15 minute. XRD technique was usee to the study the change of crystal structure while the size and morphology of the sphere were determined using SEM. DSC curve was analyzed to study reaction kinetics within the sample. SEM results indicated that emulsion process is effective on 8 minute time giving nanosphere of 482 nm with homogenous size distribution. Crystal structure of polylactide became amophous, which degree were increasing with increasing sonication time. Decrease of DSC peak intensity for longer sonication time sample support this amorphous change
