The Lumbricus rubellus earthworm consists of a group of fibrinolytic proteases or lumbrokinase that has been potentially used as a promoting antitrombotic agent. Lumbrokinase can be used as a treatment of some thrombolytic diseases such as stroke, myocardial infarction, and pulmonary embolism. In this study, we formulated protein precipitate of lumbrokinase (with standard of 150mg and high dose of 450mg) that are extracted from Indonesian earthworm powder, (L.rubellus), followed by an in vivo test of its potential antithrombotic agent using long-tail monkey (Macaca fascicularis). Earthworm powder was produced in a vacuum oven, and protein precipitate was produced by salting out precipitation using ammonium sulphate. Thus precipitation was orally administrated in monkey (Macaca fascicularis) with standard and high doses for 4 weeks. Five monkeys were divided in each control and experimental groups. Antithrombotic effects were determined based on the measurement of bodyweight changing, platelet count, prothrombin time, D-dimers level, and whole blood clot lysis time/WBCLT test. Since these results showed that there were no significant direct effects on trial animals during the treatment, we conclude that the use of formulas (standar and high doses) was safe and non toxic for further application as an orally administrated antitrombotic agent.
