An efficient DNA ectraction method is a prerequisite for molecular studies of oil palm (Elaeis Quineensisi jacq). DNA extraction from oil palm has been difficult due to its contaminants such as protein, polyphenolic coumpunds and carbobydrate. the objective of this study was to use a modified method to determine b-mercaptoethanol, a PVPP required in DNA extraction buffer to effectively inhibit polyphenol exydase, This research consisted of two stages (a) isolation of total DNA and b) DNA amplification. The isolation of total DNA included DNA extraction and purification. Young leaves were used and harvested from hybrida parent female (D10 DX D8 D), X male (L9 T X L2 T) and female (D8 DX D8 D) X male (L9 TX L2 L). In the extraction procedure, the oxidation of polyphenolic compounds was reduced by using PVPP 2% and 2-mercaptoethanol 1% in the purification procedure, the DNA was separated from other contaminans such as protein with chloroform, isoamyl alcohol (24 : 1). The concentrations of purified DNA were measured spectropphometrically at wavelengths of 260 nm and 280 nm. the ratio between the readings at 260 nm and 280 nm (A260 / A280 nm) provided an wstimate of the purity the nucleic acid, the suitability of DNA preparation for RAPD anaysis were tested with random primers 10-mer from Operon USA (OPc-07 , OPH-03 and OPH-09). The most succesful approach to obtain DNA from hybride parent suitable for RAPD analysis used the modified method of Orozco Castillo et al (1994).
