Proteases are include hydrolyses enzyme and very important, because it hydrolyzed peptides. Bond. In this research, Aspergillus oryzae was treated with the well-known mutagen N-metil-N'-nitro-N-nitrosoguanidin. (NTG), proteases enzyme source from wild type and the mutants of A. oryzae. Enzyme production with submerged fermentation method, isolation enzyme with filtered and centrifuge method, enzyme activity with Bergmeyer and Grassl method and protein concen1rat1on with Lowry method. 12 colonies survival at level 5% in a mutagenesis. The colonies are transferred on Minimal Agar, and the 7 colonies mutant resulted, the continuous screening the colonies are transfer on Milk Agar, and 5 colonies formation of clear halos near a colony, indicated produced proteases enzyme. Specific activity of mutant MAF-l, MAF-3 and MAF-4 are 2.5; 3; and 7 folds of wild type specific activity.
