Protein variants of the lymphocyte homing receptor CD44 (CD44v) are expressed in various cancer types. CD44v playa crucial role during tumor progression and formation of metastases. Immunohistochemistry methods was performed using monoclonal antibody CD44v6 to detect CD44 expression on early stage of breast cancer and the metastases on lymph node. The samples were taken from hospitalized patients in Surgery Department, Dr M Jamil hospital Padang. Tumor and lymph node tissue from 20 patients with early stages of breast cancer were obtained from surgical resection speciment. Based on TNM Classification, tumor size is classified to T1 (10%), T2 (55%) and T3 (35%1 of all casses. Whereas lymph node enlargement is classified to NO {20%j, N1 (70%) and N2 (5%) of all casses. In our microscopic investigation using indirect immunoperoxidase technique on paraffin-embeded tissue sections, we found that the percentages of CD44 expression was significant with tumor size. On the palpable lymph node (Nl & N2) the positively stained cells found in 80% C0.sses, whereas on the unplayable lymph node (NO) in 60% casses. This study show that CD44 may playa role during progression and metastases of breast cancer
