In Vitro Characterization of Pseudomonas sp. SL1.1 as a Candidate Aquaculture Probiotic (presented at the 11th AFOB Regional Symposium 2019)

Abstract
Aquatic environments possess a wide range of habitats that exhibit an enormous pool of microbial biodiversity, in which novel natural products can be discovered. Aquaculture bacteria could applied as probiotic agent in aquaculture ecosystem which is adopted from human and veterinary uses to maintain intestinal balance and biocontrol agent. Probiotic would be promising approachment to threat aquaculture microbial related disease without lead to resistance problem. Pseudomonas sp. SL1.1 had been isolated from Sawer Rahmat Waterfall, West Java, Indonesia. Candidate probiotic was further characterized using in vitro assays addressing antagonistic assay against Vibrio vulnificus and Aeromonas hydrophila, antibiotic susceptibility, determination of the isolate viability in feed, and examination the interaction on different culture conditions to evaluate probiotic activity. Cell-free supernatant of Pseudomonas sp. SL1.1 showed antimicrobial activity against V. vulnificus. Ampicillin and Novobiocin showed the weakest activity against Pseudomonas sp. SL1.1 isolate. Pseudomonas sp. SL1.1 could be applied as probiotic agent in water conditioning, co-culture, or treatment against V. vulnificus. Probiotic activity towards A. hydrophila showed only in co-culture and treatment. Nevertheless, Pseudomonas sp. SL1.1 was not withstand to drying and storage conditions into feed without any cryoprotectant. Further studies on survival of encapsulated isolate in feed producing are needed for better application.
