Abstract
Rosa sp. is a flowering ornamental plant with high economic value. In vitro culture technique could be used as an alternative method for rose propagation because it could generate a large number of plants in a given time if compared to conventional propagation. Furthermore, rose micropropagules could be used for in vitro flowering or acclimatization. Micropropagation of Rosa hybrida cv. Mawar Merah Besar was done using single node explants. First, explants were sterilized by Bayclin 20% and HgCl2 0.2%. The explants were cultured on solid Murashige and Skoog (MS) medium with 30 g/L sucrose and supplemented with different plant growth regulators combination such as BAP (Benzylaminopurine), NAA (a-Naphthaleneacetic acid) and TDZ (Thidiazuron).Rose culture was subcultured every 45 days. The best medium composition for growth and micropropagation of Rosa hybrida cv. Mawar Merah Besar was MS supplemented with 3 ppm BAP (significantly different at p<0.05 using Kruskal-Wallis). It was shown by the increased number of shoot formation, and shoot length within the subsequent medium composition. The best survival rate was attained when the rose culture were initiated and subcultured in the same medium containing 3 ppm BAP. However, MS supplemented with TDZ induced stunted and stressed shoot culture.
