Abstract
Drug transporter alteration has been detected in non-alcoholic fatty liver disease (NAFLD) condition. Hepatic multidrug resistance-associated protein 2 (Mrp2), an important transporter for biliary elimination, was suspected as one of the drug transporters that altered due NAFLD condition. Therefore, Mrp2 under conditions of NAFLD should be evaluated for any possible expression and activity changes. This study analyzed the expression and activity of hepatic Mrp2 based on the in vivo biliary excretion of mycophenolic acid (MPA) and mycophenolic acid-7-O-glucuronide (MPAG) as Mrp2 substrate in orotic acid-induced NAFLD rats (OA group) compared with control group. The pharmacokinetics of MPA and MPAG were also evaluated after intravenous (5 mg/kg) and oral (10 mg/kg) administration of MPA to NAFLD and control rats. In this research, the NAFLD model rats were made successfully due to the significant increment for its hepatic triglyceride content. Protein expression of Mrp2 seemed to be same in control and OA groups. There were also no significant changes in protein binding and biliary excretion analysis for MPA and MPAG between control and OA groups. However, there was an increment trend of MPAG biliary excretion in OA group and some pharmacokinetic parameters seemed to be change significantly in intravenous and oral study data. These results indicated that NAFLD condition in rat could change the activity of Mrp2. Nevertheless, it needs more investigation to know the detail mechanism for its changes and affects.
