Abstract
Mount Tangkuban Perahu is one of active volcano In Indonesia. It consists of four crater, Ratu crater (the biggest one), Domas crater, Upas crater, and Baru crater. Amoung those crater, Domas crater is the most accessible. People can get closer to observe the hotspring. To study microbial community in Domas crater, samples from hotspring, and soil around the hotspring were collected. DNA were extracted directy from environmental samples, followed by 16S-rRNA gene amplification. PCR products were purified and cloned into T-vector plasmid. Amplified Ribosomal DNA Restriction Analysis (ARDRA) was carried out using BstUI, HhaI, and RsaI restriction enzyme. Three kinds of samples, including soil, hot water, and mud samples from the crater were analyzed. A total of 60 clones with proportion of 20 clones for soil sample, 20 clones for hot water, and 20 clones for mud sample were constructed. There were 19 phylotypes for soil, 20 phylotypes for hot water, and 14 phylotypes for mud sample were obtained. All of three samples possessed a great number of rare and unique phylotypes, which presummably indicated that the level of bacterial diversity is very high. This study opens further research to explore microbial community in Domas crater and another crater in Mount Tangkuban Perahu.
