Abstract
Background: Aging is a natural process that happen to all living creatures. The impact of Aging process are decreasing cell regeneration, decreasing hormone level in the body and decreasing wound healing. Wound healing was influenced by many factors such as hormone, age, and nutrition. Estrogen was one of the hormones that affected wound healing. The decreasing of estrogen on skin can affect extracellular matrix such as elastin and collagen fibers. Selective Estrogen Receptor Modulator (SERM) is one of the alternative medication to prevent hormonal problems. Many studies stated that SERM have less side effects compared to Estrogen Replacement Theraphy (ERT). SERM works by binding esrogen receptor and works specifically on the target cell, thus the other side effects can be avoided.
Methods: This study is a post-test control only. Twenty eight female Wistar rats were divided into 4 groups. K7 and K14 were control groups, P7 and P14 were treatment groups. The duration of this study are 7 days and 14 days. All rats were shaved in dorsal area in 2x2 cm, then a full thickness incision in 1 cm of length were made. Control groups were treated with topical erythromycin 2% on the wound once a day. Treatment groups were treated with topical mixed of erythromycin and tamoxifen 10 mg on the wound once a day. Samples were taken after 7 days and 14 days. The rate of elastin and collagen fibers will be examined by microscopic examination using 1 high power field (HPF) in percentage system.
Results: Collagen mean percentage for group K7 is 45.27%, P7 is 58.30%, K14 is 50.45%, and P14 is 60.42%. Elastin mean percentage for group K7 is 0.96%, P7 is 1.34%, K14 is 1.01%, and P14 is 1.37%. The results show increasing value in treatment groups for both elastin and collagen. The obtained data were then analyzed using One Way ANOVA, and the result shows p = 0.00 (p<0.05 for both elastin and collagen groups). There is a significant difference between groups.
Conclusion: The elastin and collagen mean percentage in treatment groups (P7 and P14) are higher than control groups (K7 and K14). In conclusion, topical SERM tamoxifen increased the quantity of elastin and collagen in menopausal Wistar rat wounds
