Abstract
Background: CYP2B6 gene carries slow metabolizer characteristic towards drugs that are metabolized by CYP2B6 enzyme and are widely used such as anti malarial, anti retroviral, tamoxifen, bupropion, etc. This condition increases toxicity risks for the patient and changes the therapeutic outcomes. This research was conducted because there was no research about CYP2B6 prevalence in Indonesian population. Timorian is chosen because East Indonesia is still endemic to malaria.
Methodology: Polymerase Chain Reaction and Restriction Fragment Length Polymorphism (PCR-RFLP) are used to determine prevalence of SNP 516G>T and 785A>G from 96 Timorian patients from South Amanuban District. DNA fragments are separated using electrophoresis and gel documentation is used for DNA visualization. Microsoft excel is used for all statistical analysis.
Results: Demographic data shows 35 male subjects dan 61 female subjects with mean age 39.9 years old for male subject and 49.9 years old for female subjects. Allelic prevalence of 516G>T and 785A>G in Timorian are 56.8% and 46.9%. The highest genotype prevalence, which is *6/*6, is 22.9%, followed by *1/*6, which is 5.7%. Genotype prevalence of other variants are *1/*4, *1/*9, *4/*4, *4/*6, *6/*9, and *9/*9 , which are 4.2%, 6.3%, 1%, 5.2%, 10.4%, and 7.3% respectively. Allelic prevalence of CYP2B6*6 is 41.1%. Prevalence of other type of alleles, CYP2B6*4 and CYP2B6*9 are 5.7% and 15.6%.
Conclusion: The prevalence of CYP2B6*6 in Timorian is similar to West African population and smaller than Papua New Guinea population. The highest genotype prevalence of CYP2B6*6 is *6/*6 and followed by *1/*6 and has the same pattern as Papua New Guinea population. High prevalence of CYP2B6*6 can alter the therapeutic outcomes of antimalarial in Timorian.
