Abstract
Backgrounds. Diabetes mellitus is a progressive metabolic disease and has become one of the most challenging public health problems in the 21st century. Presently, diabetes is managed or controlled using antidiabetic agents. However, all the antidiabetic agents only can alleviate symptoms and also not devoid of adverse effects. These conditions encourage researches in finding new therapy that could avoid progressive effect of the disease by maintaining pancreatic beta cell.
Objectives. The present investigation was aimed to assess the effect of okra extract in lowering blood glucose and protecting pancreatic beta cells against cell damage in alloxan-induced diabetic rats.
Methods. The method of this study included 25 male Sprague Dawley rats which were injected with high-doses alloxan in order to mimic condition of type 2 diabetes mellitus. Samples were grouped randomly into 5 groups which were given 100 , 200 , 400 extract, and placebo (control group) respectively for 21 days. Blood glucose levels were measured before intervention with okra extract and repeated every week after intervention. After 21 days, pancreatic tissues were taken and stained with monoclonal insulin antibody in order to count number of pancreatic beta cells with 400x magnification. Statistical evaluation was done using one way analysis of variance (ANOVA) followed by Turkey’s test to analyze the differences between the groups. The data obtained from blood glucose level test were analyzed using repeated ANOVA.
Results: Significant blood glucose reduction were started to shown by all groups that received okra extract after two weeks of intervention. This study also showed
significant increment of pancreatic beta cells number in all groups with okra extract compared to positive control.
Conclusions. A.esculentus extract have beneficial effects on the reduction of blood glucose level and protecting pancreatic beta cells against cell damage in alloxaninduced diabetic rats.
