Abstract
Background: Escherichia coli (E. coli) is a Gram negative bacteria that can causes various disease such as diarrhea, urinary tract infection, septic shock, and meningitis. E. coli have a high level of resistance to various antibiotic, making it difficult to cure diseases caused by these bacteria. To overcome the E. coli bacterial resistance to antibiotics, the use of herbal remedies may be an alternative for treatment. Indonesia has a diversity of medicinal plants, but not many scientific studies that prove the effectiveness although it has been widely used as a traditional medicine in society. In some studies, Piper crocatum leaf extract (one of the red betle species in Indonesia) proved to have the antibacterial effect against Gram-positive bacteria (Staphylococcus aureus) and Gram-negative bacteria (E. coli and Steptococcus mutans). Therefore, this experiment used Piper decumanum L., another red betle species which is also one of Indonesia's traditional medicinal plants, which have never been studied before. Methods: This is an in vitro experimental research. By employing maceration process (using 96% methanol as solvent), the red betle leaves is extracted. The process takes 3 days to complete. The antibacterial testing is carried out with dilution method continued with pour plate method. The research uses concentration of 50%, 25%, 12,5%, 6,25%, 3,13%, and 1,56%. Results: Even the result of statistic test with Kruskal-Wallis show p>0.05, the experimental results show that the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) at the extract concentration of 25%. Conclusions: The methanol extract of red betle leaf might have a capability as an antibacterial agent against Escherichia coli.
