Background: Pterygium is one of the commonest conditions encountered in temperate regions. UVB exposure is associated with the etiology pterygium causing mutations in tumor suppressor gene, p53. Discovery over expression of p53 gene mutations in pterygium and tumor characteristics such as the treatment modality could be an anticancer agent. Five fluorouracil (5FU) is the agent of the group antagonist pyrimidine, inhibits the proliferation of fibroblasts and affects replication of S phase cells, acting on RNA. While mitomicin C (MMC) derived from Streptomices caespitosus, has a role to inhibit fibroblast proliferation by blocking DNA replication (cell proliferation phase of G and S), mitosis and protein synthesis. Chemotherapy agents such as mitomicin C and 5 Fluorouracil was used as a adjuvant therapy and aim to reduce the recurrence rate of pterygium have never studied its effect on the mutant-type p53 expression through Imunohistochemistry examination. Objective: This study aims to determine the expression ratio of p53 in primary progressive pterygium after sub-conjunctiva injection of Mitomicin C and 5 fluorouracil. Methods: Quasi experimental design with posttest only, performed at Dr. Sardjito Hospital and networks are regrouped into 2 groups with injection treatment subconjunctival mitomicin C or 5-¬FU 1 week before surgery. Tissue stored in buffered formalin, staining with the monoclonal antibody DO-7, mutant p53 expression is calculated per 100 cells in 5 fields of view, 400 x magnifications. Result and Conclusion: There were 53 subjects, 27 specimens in MMC group and 26 specimens of group 5 FU. Not found significant differences in the characteristics of Subjects. In this study, the expression of p53 of MMC groups was 5.71 +1.83, whereas the rates of 5FU group p53 expression was 10.29 + 4.54 (p = 0.000). Variable age showed a significant correlation of 0.29 and the average influence p53 expression. The mean expression of p53 in mitomicin C injection group is lower than 5 Fluorouracil groups and significant different statistically.
