Abstract
Introduction
Distant metastasis in breast cancer is attributable to a severe decrease in the five-year survival rate of patients from approximately 60% to 20%. Metastasis may be initiated by cellular hypoxia, which stimulates the migration of neoplastic cells towards distant sites. PDIA4 (Protein Disulphide-Isomerase family A, member 4) is thought to be a tumor biomarker of high sensitivity and specificity towards metastasized breast cancer. PDIA4 is a hypoxic stress protein that increases the cancerous cell’s resistance against hypoxia. The aim of this study is to find a correlation between the amount of PDIA4 within cancerous cells and the probability of distant metastasis in breast cancer for the purpose of finding a non-invasive and accurate means of diagnosing metastasis.
Method
mRNA samples from 21 metastasized and 18 non-metastasized breast cancer tissues were amplified for PDIA4 using reverse-transcription, qPCR and analyzed using Comparative CT Livak method. The Human ACTB (Beta-actin) gene was used as control.
Results
PDIA4 mRNA expression ratio is analyzed using the Livak method.Shapiro-Wilktest shows that the data distribution is abnormal (p<0,05). The mean value of PDIA4 mRNA expression ratio for the metastatic cell group is 4.72 and for the non-metastatic cell group is 3203.98. Based on Mann-Whitney correlation method, there is significance difference of mRNA PDIA4 expressions between the two groups (p<0.001). Result of Spearman correlation test is p=0.000 and R=-0.651 which mean there is significance strong correlation between mRNA PDIA4 expresssions towards metastasis status.
Conclusion
Metastatic breast cancer cells express a statistically significant lower level of PDIA4 mRNA when compared to non-metastatic breast cancer cells and influence the level of mRNA PDIA4. PDIA4 continues to be a promising biomarker for early detection of breast cancer metastasis.
