Abstract
Ice nucleation-active (INA) bacteria is environmental bacteria which have capability to induce ice formation at temperature warmer than -10 °C . While common in temperate regions of the world, little is known about the presence and distribution of INA bacteria from tropical regions. Therefore, studies on INA bacteria from tropical areas, especially Indonesia, and their role in atmospheric processes leading to rain were needed. Rain-water samples and air were collected from several locations near Jakarta, Indonesia for one year period of time. In this study, twenty-six isolates were classified as class A (active between -2 and -5 °C), while 2 isolates as class B (active between -5 and -8 °C) and 77 isolates were categorized as class C (active only below -8 °C). This bacteria well known as plant pathogenic their potential in causing frost injury at plants. We performed eighteen isolates of INA bacteria recovered from rainwater collected in Bandung, East Jakarta, Bogor, and West Jakarta. We characterized the properties of this isolates using I6S rRNA gene PCR amplification and continued with DNA sequencing analysis, antibiotic resistance assay, biofilm formation assay, EPS biosynthesis, as well as quorum sensing and quorum quenching formation assays. We performed that 61.11% isolates were similar to Pantoea ananatis with homology range from 98% to 99%, and 11.11% of the isolates were similar to Pseudomonas lurida with homology range from 97% to 100%, while 11.11% isolates were similar to Pantoea agglomerans with 99% homology, and the rest showed 99% homology with Pseudomonas mosselii, 98% homology with Cronobacter sakazakii, and 99% homology with Klebsiella pneumoniae. The presence of INA bacteria in rain-water and air might plays an important role in nucleation process which is required for rainfall induction even in tropical areas.
