Abstract
Vibrio cholerae is well recognized as the causative agent of cholera, an acute intestinal infection characterized by watery stools which leads to dehydration and to death in some cases. V. cholerae is a natural inhabitant of the aquatic environment. Inadequate safety measures in drinking water supplies, poor sanitation and hygiene can increase the risk of cholera outbreaks. There is limited study on these bacteria from ice and beverages that are popularly vended and consumed in Jakarta, Indonesia. In this study we have enumerated and detected V. cholerae from ice and beverages collected from several areas comprising five different regions of Jakarta. The prevalence of V. cholerae in both ice and beverages was determined by using three-tube MPN method, and ranged between 11000 MPN/ml. We have also screened the presence of regulatory and virulence genes using multiplex PCR. From this study we found that 33 of 110 samples were contaminated by V. cholerae, 21 of which were from ice samples. A total of 88 isolates of V. cholerae yielded positive for toxR gene, alone or in combination with other gene(s) of which 57%, 25% and 19% were hlyA, ompU and ctxA, respectively. The detection and isolation of potentially harmful V. cholerae from ice and beverages in Jakarta may indicate that these products are not completely free from choleragenic vibrios.
