Since the first workshop was held by this group in 1989, there has been a great expansion of information on the structure and possible origins of anti-DNA antibodies. Study of these characteristic SLE autoantibodies has increasingly progressed to the molecular level. Participants in this 1996 workshop reported on: VH domain sequencing and cloning, expression and mutagenesis of Fv or Fab fragments; computer modelling and crystallography of antibody and DNA-antibody complexes, and regulated expression of potentially autoreactive antibodies in transgenic mice. Molecular analysis extended to antigen presentation, T cell activation and pathogenesis. Thus, a striking feature of this 1996 workshop is that discussion of the biological processes has become more firmly based on knowledge of the structure of participating reactants and their coding genes.
