Isi artikelThe idiotypic network hypothesis, as proposed by Jeme,’ predicts that interactions between antibodies via idiotopes (types) and between T cells and B cells via idiotype-specific receptors are critical in the regulation of the immune system. Cross-reactive idiotypes (CRI) on anti-DNA antibodies have been identified in both murine models of lupus2-5 and in human lupus,6-9 and investigations into the role of idiotypic regulation in the pathogenesis of SLE have demonstrated their importance. Immunization with an immunoglobulin light chain expressing a CRI associated with murine anti-DNA antibodies induced autoantibody production in normal mice.’° Induction of an experimental disease resembling murine SLE was achieved when normal mice were immunized with a human anti-DNA antibody carrying the 16/6 idiotype (Id).&dquo; Anti-Id antibody has been shown to modulate the production of anti-DNA antibody both in vitro’2 and in vivo’3,’a and both Id-specific CD4 + helper’s°’6 and CD8 + suppressor cells&dquo; appear to be involved.
