The demonstration of antigen selected mutations in the CDR (complementarity determining regions) of monoclonal anti-dsDNA antibodies in lupus-prone mice and humans with SLE are clear indications that these antibodies are antigen-driven and T cell dependent. The origin and nature of possible antigens being responsible for the spontaneous production of anti-DNA antibodies, however, is still an unsolved problem. Since native human dsDNA per se is not immunogenic, several concepts regarding the induction of anti-dsDNA antibodies have been developed, including non selective polyclonal B cell activation, selective B cell stimulation or stimulation by crossreactive antigens.
