Advances in our understanding of the genetic control of IgH chain gene usage in production of anti-DNA antibody were reported using three representative murine models: (i) a spontaneous murine lupus model (NZB x NZW) Fl female mice; (ii) a transgenic mouse model carrying a numan IgH minilocus (with a VH of a human anti-DNA antibody); and (iii) an anti- DNA antibody variable region gene targeted mouse model. Each model has its advantage to address certain aspects of structural characteristics and regulation of antibodies to dsDNA. Lessons learned from each model are summarized in the following.
