This article reviews recent discoveries of the molecular basis of several primary immunodeficiency diseases, including four x-linked recessive and seven autosomal recessive conditions. Other conditions for which new information is available, such as genetic mapping, as well as new immunodeficiency syndromes also are discussed. Advances in the therapy of immunodeficiency diseases are reviewed, including the development of generally safe and effective intravenous immunoglobin preparations and T cell depletion techniques that permit the use of non-HLA-identical bone marrow donors for immunologic reconstitution.
