A gene encoding an extracellular lipase was identified in Staphylococcus kloosii EP2.22. Full length of lipase gene was obtained by Genome Walking Method. Using this method, first sequence 332 bp could become a clue for upstream and downstream amplification. DNA sequencing and gene cloning showed an open reading frame of 2322 bp, which encodes lipase enzyme of 774 amino acids. The catalytic triad residues consists of Ser511, Asp703, and His742. Homology alignment was employed to analyze sequence function relationships with others Staphylococcal lipases. In amino acid sequence, S.kloosii EP2.22 lipase only showed 52.7% pairwise identity with S. warneri lipase. |
