Abstract
Vibrio cholerae is the causative agent of the endemic and epidemic diarrheal cholera in areas with poor sanitation and the disease occurs sporadically or as limited outbreaks in many developing countries. The pathogenesis of cholera is a complex process, including the ability to adhere to, and a number of factors to colonize the epithelium of the small intestine and produce enterotoxins that disrupt ion transport. Sample of beverages were collected from some areas in Jakarta for MPN analysis; the three-tube MPN method was used. Fifty beverages samples were inoculated into Alkaline Peptone Water (APW) with serial dilution and detection of virulence genes were done using Multiplex PCR. The highest maximum number for the MPN result reached >11000 and the highest average number is 4070 both from sample B.NJ.5 and B.NJ.6. The lowest maximum MPN number (<30 MPN/ml) was found in eleven samples. A total of 50 beverage samples were examined for the virulence gene detection. Twelve (24%) samples were positive for toxR PCR detection with amplicon size of 779 bp, 58.3% of the positive samples were from East Jakarta, 25% were from North Jakarta, and 16.7% were from South Jakarta, while samples from West and Central Jakarta showed negative result in toxR detection. From the Multiplex PCR, we found 11 positive isolates for toxR and hlyA detection. Combination of MPN and Multiplex PCR can be used for prevalence and detection of virulence and regulatory genes of V. cholerae from beverages in Jakarta.
