Laboratory studies have shown the existence of certain strains of bacteria in oil fields and brines. This investigation is concerned with studying the effect of indigenous bacterial activities on the alteration of crude oil properties. Ten bacterial strains were isolated from crude oil and brine samples of an oil reservoir in Kalimantan, Indonesia by using Stone Mineral Salt Solution (SMSSe) medium supplemented with 0.1% (w/v) yeast extract. These results revealed that all of the bacterial strains isolated were capable of utilizing crude oil as their sole source of carbon and energy for growth. The strains that showed the highest growth in the screening medium (brine supplemented with 2% (v/v) molasses, 2% (v/v) crude oil, and 0.1% (w/v) urea) in order of ranking were identified as Flavimonas oryzihabitans, Amphibacillus xylanus, Bacillus polymyxa, B. macerans, B. stearothermophillus, and Bacillus sp., respectively. All strains were also able to grow at high temperatures (50-90°C). There were physical and chemical changes (i.e.: viscosity, interfacial tension, and oil swelling) in the crude oil as well as solution acidification of the bacterial cultures after 4 days of incubation. These results suggest that the bacterial strains have produced bioproducts. This study therefore demonstrates the potential of the bacterial strains on microbial-enchanced-oil-recovery applications.
