Abstract
Direct organogenesis from leaf sections of two lines of Jatropha curcas L. was obtained. Shoot buds regeneration from leaf explants were formed on an initial basal medium Murashige and Skoog (MS) supplemented with different concentration of Thidiazuron (TDZ) and 6-Benzylaminopurine (BA) in combination with Indole-3- butyric Acid (IBA). All treatments were subjected to MS medium with 2 ppm BA + 0.5 ppm IRA for shoot proliferation, further to MS medium containing 0.5 ppm BA for shoot elongation. Both types of cytokinins tested were capable of inducing direct organogenesis from J. curcas leaf sections. TDZ was observed to favor the induction of direct organogenesis which associated mostly with leaf veins. On the other hand, the combination of BA and IBA favored the induction of indirect organogenesis via callus. TDZ although performed better during initiation failed to produce shoots which might be correlated with habituation and plant growth regulator types or concentration compatibility between the induction and proliferation medium in the explants. JC-7 line was found to be more responsive in medium containing 3 ppm BA + 1 ppm IBA while the other line (JC-8) was best in medium containing higher concentration of BA (6 ppm BA + 1 ppm IRA)
