Good parameter estimation methods are essential in modeling of biochemical reaction systems. We have developed a parameter estimation method for biochemical reaction systems based on the Levenberg-Marquardt method for non-linear least squares problems. The method has also been extended to take constraints on parameters into account using barrier functions. Performance and convergence properties of this local optimization method are compared with a global optimization method based on a simulated annealing variant of the Nelder-Mead non-linear simplex method. The comparison is carried out by using three different nonlinear biochemical dynamic models as benchmarks. |
