Lipase is one of important enzyme classes that is commercially produced by microbes. Bacillus brevis BSWt 2(1) is an endophytic bacterium isolated from the mesocarp of palm fruits. It could produce unique lipase in Kouker and Jaeger’s medium containing olive oil. The enzyme was very stable at high temperature, however, the enzyme activity was low. Optimization of the enzyme production was carried out with modification of the media compositions. Those were variation of olive oil consentration (1.0 and 2.5 %), variation of carbon source composition (1.0 % olive oil, 0.5 % Tween 80, 1.0 % olive oil and 0.5 % Tween 80, and 0.5 % olive oil and 0.5 % Tween 80), and sources of nitrogen (mixture of polypeptone, yeast and beef extracts; urea; monosodium glutamate (MSG); and soybean meal). The optimum production was obtained at the consentration of 1.0 % olive oil and 0.5 % Tween 80, and MSG as the nitrogen source at the four days of incubation time. |
