Development of the fruit is needed to push up by govermer or consumer. Supplying fruit seedling by conventional methot like stack, grafting or seedling in large quantity is difficult because of time and uniformity. In vitro culture is one of alternative technology to produce large number of seedling in short time and uniformity. The experiment was conducted at laboratory and green house of Biology Cell and Tissue culture Division of BB Biogen Bogor in 2004. There were some steps of experiment : Increasing shoot multiplication, root inducting, and aclimatization. Media for shoot multiplication were Woody plant medium (WPM) combined with kinetin (1 and 3 mg/l) + GA3 (3 and 5 mg/l). Arginine 100 mg/l was added in the medium because of slow growth and mutiplication of the shoot. The other media also were tried in high dose WPM (W) combined with BA (B) and thidiazuron W B10mg/l; W B10mg/l thi5mg/l; W B10mg/l thi10mg/l; W thi10mg/l; dan W thi10mg/l B5mg/l). Root inducting media were WPM (1, ½) + IBA ( 0, 1, 3, 5 mg/l); Planlet with fine root was aclimatization in green house. Observation was cunducted on shoot inisiasion, number of shoot multiplication, shoot height, root inisiation, number of root, visual observation and succses of aclimatization. Result of experiment was showed that WPM + Kinetin 1 mg/l + GA3 5 mg/l + Arginin 100 mg/l increase shoot multiplication and root formation caused planlets were ready to aclimatization. Combination medium between basic medium WPM and BAP also thidiazuron with hight dose were not increase shoot mulyiplication and root initiation of salak condet.
