Abstract
There are 3 phases during the development of soybean suspension cell culture. Those phases are
leaf, callii and suspension cells. During the development process, the respiration through cytochrome
(COX) pathway and alternative (AOX) pathway was monitored and compared. Respiration were measured
at tissue level in which respiration of the whole tissue was monitored and at mitochondrial level in which
the respiration of the isolated mitochondria was monitored.
Whole cells respiration through cytochrome pathway (COX) and alternative (AOX) pathway were
also monitored during 7 days in culture which is a period for subsequent subculture.
In this experiment cytochrome pathway respiration is more dominant than alternative pathway
respiration at leaf and suspension cell phases whereas the alternative pathway respiration is more dominant
at the suspensiion cell phase. This indicates that under a certain condition respiration through the
alternative pathway can be the main operating pathway of the mitochondrial electron transport chain.
Whole cell respiration during a week culture period showed a peak at 3 days after subculture (DAS)
whereas for alternative pathway the peak was reached at 2 days after subculture (DAS)
Immunoblot of AOX proteins from several tissues of soybean indicates that there were 2 isoforms of
alternative oxidase proteins obtained from mitochondria isolated from leaf and cotyledons whereas
immunnoblot from mitochondria isolated from root, callii and suspension cells showed only 1 isoform of
AOX proteins. This experiment indicates that eventhough immunoblot showed the abundance of the AOX
proteins, the respiration through AOX pathway was not correlated.
