Abstract
Zinc concentrations found within the retina are relatively high compared to the rest of the body. In addition, zinc-deficient diets and mutated proteins (involved in the signal transduction mechanism found within rod cells) have been linked to photoreceptor degeneration and blindness. The objective of this study was to establish a reconstituted system to investigate whether the presence of zinc had a direct effect upon rhodopsin phosphorylation in the presence and absence of various ROS components. Using rod outer segments, previous studies showed an increase in rhodopsin phosphorylation in the presence of zinc. In this thesis work, a reconstituted system was established, using purified rhodopsin and rhodopsin kinase from bovine retinas. In this in vitro system, rhodopsin kinase phosphorylates vesicularized rhodopsin in a light-dependent manner. Further research will be necessary to establish the proper conditions for zinc enhanced rhodopsin phosphorylation.
