Differentiation of pre-adipocytes into adipocytes, is a complex process that involves coordination of expression of specific genes and proteins associated with each stage of differentiation. This process is regulated by several signaling pathways. CCAA T element-binding proteins (C/EBP) a and f3 and sterol response element-binding protein 1 (ADD1/SREBP1) are active during the early stages of the differentiation process and induce the expression and/or activity of the peroxisome proliferator-activated receptor r (PPARn, a pivotal coordinator of adipocyte differentiation. Recently, many biological effects of black tea have been reported and these effects may be partially related to its antioxidant properties. The recent study showed that black tea fIavonoids inhibited adipocyte differentiation. The aim of this study was to investigate the role of theaflavin, a fIavonoids in black tea, in inhibiting adipocyte differentiation through inhibition of PPARr expression. Preadipocytes were incubated in media containing standard-differentiated stimulator: Insulin, dexamethasone, IBMX and FBS. After 24 hours, preadipocytes culture media was added with theaflavin 50, 100, or 200 j.iM. The cells were harvested after TF incubation for 24 hours and then the expression of PPARrwas identified by immunocytochemistry and Western blot. The number of differentiation of preadipocyte decreased after incubation with TF for 24 hours. Cell differentiation significantly decreased following incubation with TF100 j.iM (24.08:t16.09) and TF200 j.iM (28.05:t6.24) compared to the control (65.84'1:16.92) and TF50 JIM (64.93'1:10.46) treatments. The number of undifferentiated cells increased significantly in groups treated with TF 100 JIM (75.92:t10.46) and TF200 j.iM (71.95:t6.24) (p<0.05). The results of immunocytochemistry and Western blot showed that PPARr expression decrease in TF200 j.iM treatment. Theaflavin polyphenol of black tea could decrease preadipocyte differentiation which may be through the inhibition of PPARrexpression.
